According to John Campbell’s Single Shot book, yes there was a 25-21 chambering although he noted it to be very rare.
I’m sure Bert "High Wall" Hartman will chime in soon.

Robert,
Winchester did make Single Shot rifles in the 25-21 Stevens cartridge, but most of them were low-wall rifles. The actual number as surveyed was just (95) of them.
